Some Brain Dysfunctions induced by Acrylamide and possible mitigating role of Alpha-lipoic acid in adult rats

Abstract

The current study was conducted to find out the neurotoxic effect of the Acrylamide (ACR) and the role of Alpha lipioc acid (ALA) to ameliorate the ACR neurotoxicity.

Fifteen adult male rats were subjected in this study, and assigned randomly into three equal groups. Control group (CCG), were had tab water for 45 days by gavage needle, Acrylamide group (ACG), rat in this group had acrylamide (ACR) 5mg/kg B.W by gavage needle for 45 days and Acrylamide and Alpha Lipioc group (ALG), rats had acrylamide (ACR) 5mg/kg B.W. and Alpha lipioc acid (ALA) 100mg/kg B.W. for 35 days orally as well. At the end of study, the behavioural testing was performed and brains were harvested for histopathological examination.

The current studies showed the oral exposure to ACR impaired rats neurobahaviors, while treatment of ALA retrieved the normal behaviours.

Moreover, histopathological examination of brain sections animal exposed to ACR

shows  microglial cells aggregation in one side of blood vessels in parenchyma and inflammatory cells  around  and in congested blood vessels, furtyhermore, proliferation of  microglial cells with Alzheimer type II astrocyte were notice as well. However, the sections of ALG showed ameliorative effect in this regard.

In Conclusion Acrylamide impaired some brain activities and structure, and alpha lipoic acid shielded the brain and restore the normal function, and structure.
